The Italians need a win at the Stadio Olimpico to maintain their hopes of reaching the next round, while defeat in Rome would dent their chances of progressing.
"This game is very important for us," Hernanes said. "We want to go as far as possible in this competition.
"We are playing at home and our aim is to take a win."
Vladimir Petkovic's side lie second in Group J with five points after three games, two points adrift of pool leaders Trabzonspor and one point clear of Apollon.
Lazio were held to a goalless draw at Apollon two weeks ago and Hernanes knows his side will need to give something extra to claim three points.
"Apollon have proved to be a good team," he said. "We will have to be very focused.
"They will try to frustrate us but we just have to focus on our game plan and make the most of our chances."
Lazio expect to bounce back from their 1-0 loss to Genoa at the weekend which saw their unbeaten run at home come to an end.
The result also left the Biancoceleste seventh in Serie A.
Apollon go into the encounter on the back of Saturday's 2-0 triumph over APOEL which kept them at the top of the Cypriot First Division.
The result extended Apollon's winning run in the domestic competition to six games.
Apollon coach Christakis Christoforous will be hoping his side can show the same balance they displayed against the Italians last month in Cyprus.
"Against Lazio, my players exceeded themselves and they deservedly earned the point, "he said.
"We will continue to work hard for the best in this competition."
