It is likely Sir Alex Ferguson will rotate his squad for the visit of the Canaries, but Hughton is under no illusions over the magnitude of the task which awaits his men when they run out at Old Trafford.
"It is a massive game they have on Tuesday. I think it will affect the line-up, but I don't think it will affect the quality of the line-up," said Hughton, whose side recorded a first win in 10 league games when they came from behind to beat Everton 2-1 at Carrow Road last weekend.
"Irrespective of the team they put out, we know it is going to be one of high quality.
"At this moment, they are the team who have the best quality squad in the division, there is no doubt about that.
"There is nobody who manoeuvres his squad any better than Sir Alex."
Hughton added: "We were on the back of a few draws but we managed to get that win against a very good Everton team. Scoring so late shows the resilience and determination we had to get three points.
"We hope we have benefited from it but certainly so going into the game at Old Trafford, because it has given us a good feel this week."
Norwich recorded a shock 1-0 win over United at Carrow Road back in November, but Hughton accepts an historic double over the Red Devils is a long shot.
"United have only lost one game at home this season so the magnitude of the task is still there, but we are certainly better going into it for the result of last weekend," he said.
"Beating United at home was one of our best performances of the season, and it had to be because that is the only way you can beat a Manchester United team.
"Any win at Old Trafford would have to see us play extremely well and for United to have one of their rare off-days.
"But you have to go into every game hoping the work you have done can come right and you can get that result."
Hughton added: "There will be no fear from our players, we will want to go there and play.
"However, it is very difficult to go to Old Trafford with an open formation. If you are going to go there and take them on at what they do very well, then generally there is only one outcome.
"Ultimately you want to get a result, and a draw at Old Trafford is a very good result, but it is also about a performance."
On-loan forward Kei Kamara came off the bench to head an equaliser against Everton - and is the first player from Sierra Leone to score in the Premier League.
While the Sporting Kansas City striker has become an instant cult hero among fans, Hughton is keen to keep a lid on expectations.
"Here we are keen to play it down a little bit," said Hughton.
"I understand the interest in him, but at the moment he has only made two appearances and is developing his game.
"Kei is part of a very hard working team who are starting to get results. The impact he has made has been very much for the team."
Norwich will again be without midfielder Alex Tettey because of a knee problem, while winger Andrew Surman is also still sidelined by his knee injury.
England goalkeeper John Ruddy is on target for a return at the end of March following a thigh operation.
