Christian Eriksen, Mousa Dembele and Jan Vertonghen were absent from Tottenham training ahead of Wednesday night’s clash with Barcelona at Wembley.
Vertonghen and Dembele came off at half-time at Huddersfield on Saturday while Eriksen has been carrying a groin problem.
Manager Mauricio Pochettino will give an update on the trio when he faces the media at 1pm.
Goalkeeper Hugo Lloris was back in training after a thigh injury but Dele Alli, as expected, was missing with his hamstring problem.
Spurs later confirmed all three will miss the tie and, along with Alli and Serge Aurier, will not be available until after the international break.
That also rules all five players out of Saturday’s Premier League visit of Cardiff.
Striker Harry Kane is confident the injuries will not derail Tottenham as they bid to bounce back from their defeat at Inter Milan.
“That’s the reason we have a big squad, it’s time for the next man to step in,” he said.
“Of course it’s a massive game in the group, you never want to start with two losses. At home we have to be strong if we want to progress.
“Barcelona are a fantastic side but last year we proved we can beat the big teams, especially at home.”
